First of all, I would like to note that anything to do with "shipping" does not belong here.
Because Saix/Isa was Axel/Lea's original friend, and the two of them grew apart during their time in the Organisation, I think that Axel wanted to be Roxas' "friend" more out of desperation than anything else. He knew that there was no hope in him befriending Saix again, as the two of them were too different by that point, and because Roxas was a "clean slate" with no memories of a past life, Axel seized the opportunity and decided to pretend to befriend Roxas, because befriending someone with no real past seemed to be a very easy task.
Other moments in the games (e.g. Axel saying that he would miss Roxas if he left the Organisation), despite the fact that they may seem genuine, are simply Axel becoming more desperate not to lose his replacement. He treats Roxas as an object above anything else, and he would not have befriended Roxas if Saix had stayed with him.

To me, this dialogue seems to be quite forceful on Axel's part.
